چکیده

It is estimated that at least 50 million people in the United States suff er from chronic pain conditions while an additional 25 million people suff er from acute pain [1]. Pain aff ects patient quality of life and is a major reason for healthcare utilization [2], accounting for 20% of outpatient visits and 12% of all prescriptions [3]. Nonsteroidal anti-infl ammatory drugs (NSAIDs) remain a main stay of therapy due to their effi cacy as anti-infl ammatory/anal gesic agents. In 2004, persons in the United States spent more than $2.5 billion on over-the-counter NSAIDs and fi lled more than 100 million NSAID prescriptions [4]. Worldwide, over 73,000,000 prescriptions for NSAIDs are written each year [5].

NSAIDs, gastrointestinal injury, and cytoprotection.

Gastrointestinal toxicity caused by nonsteroidal anti-inflammatory drugs (NSAIDs) is the most frequent drug side effect in the United States. NSAIDs are implicated in the development of complicated peptic ulcer disease and injury to the small bowel and colon. NSAIDs interfere with prostaglandin-mediated epithelial defense mechanisms and also cause direct epithelial toxicity. Current and future ...

Modulatory role of nitric oxide releasing NSAIDs in aging- and lipopolysaccharide-induced cognitive dysfunction in mice

Inflammatory processes play a critical role in the pathogenesis of the neurodegenerative disorders and are associated with cognitive impairments. Nonsteroidal anti-inflammatory drugs (NSAIDs) and nitric oxide releasing NSAIDs are reported to be effective in reducing the risk of developing AD or cognitive impairments. Present experiments were performed to study the possible effect of nitro napro...

Cardiovascular and gastrointestinal toxicity of selective cyclo-oxygenase-2 inhibitors in man.

It is well established that the use of traditional nonsteroidal anti-inflammatory drugs (NSAIDs) increase the vulnerability of the gastrointestinal (GI) mucosa for the development of peptic lesions and serious ulcer complications. In addition, selective and traditional NSAIDs have also been associated with increased frequency of cardiovascular toxicity, especially in susceptible patients. The o...
